Scientists from Canada and Australia studied how climate change may be affecting microbial communities by performing metagenomic and 16S rRNA gene analysis of bacteria collected from Arctic sea ice and nearby sea water. The different habitats harbored very different microbial populations. Metagenomic sequencing was performed with the Ion Torrent PGM after size selection with the Pippin Prep system.

Two stable variants of Burkholderia pseudomallei strain MSHR5848 express broadly divergent in vitro phenotypes associated with their virulence differences

Scientists from the United States Army Medical Research Institute of Infectious Diseases compared two strains of Burkholderia pseudomallei to find variants that might explain why one version was significantly more lethal than the other. They hope this work could lead to biomarkers in cases of infection. For genome analysis, they used BluePippin to size-select libraries for PacBio sequencing.

Detection of individual ploidy levels with genotyping‐by‐sequencing (GBS) analysis

Utah State University researchers developed a statistical approach to detect low-level ploidy variants using genotyping data from a sequencing workflow. The method does not require prior ploidy information to function. An evaluation was performed on Populus tremuloides, in which it successfully distinguished diploids, triploids and tetraploids. For this work, BluePippin was used to select 300 bp – 400 bp fragments prior to sequencing.

Rapid genetic and morphologic divergence between captive and wild populations of the endangered Leon Springs pupfish, Cyprinodon bovinus

Scientists studied members of two remaining populations of the endangered Leon Springs pupfish, one in a desert spring and the other in captivity. By analyzing SNPs, they found significant genetic differentiation between the populations. BluePippin was used for tight size selection.

De Novo Genome and Transcriptome Assembly of the Canadian Beaver (Castor canadensis)

Researchers in Toronto report a high-quality draft assembly of the 2.7 Gb beaver genome using SMRT Sequencing. They include an analysis of gene content, such as enrichment for genes associated with dentition and enamel deposition. The team used BluePippin to remove fragments smaller than 15 Kb before sequencing with PacBio.
